Be absolutely sure to remove those who ask to unsubscribe quickly and politely. If you do it right, you could very well get an email subscriber back, just because of good customer service. It's important to realize that these messages are opportunities. Take them as constructive criticism and opportunities to improve.
Take a look at your newsletter. Remember that your competitors are up on all the technologies and it's easier than ever to make a newsletter sharp and spiffy. Make sure yours is as fresh as it can be. The content should be up-to-date and with no broken links. Be sure the content is relevant and effective. Be sure the newsletter is friendly on the eyes and that your contact information is clearly displayed.
